- Abstract
This research investigated the effects of different media and environmental treatment (light and dark) on biomass and secondary metabolite production in callus culture of Kaemferia galanga. Environmental light had no significant effect on biomass callus. Media with sucrose 30 g/L under light condition gave the highest biomass callus. Kaemferia galanga L callus contained alkaloids, flavonoids, saponins, steroids and ethyl p-methoxycinnamate (EPMC). Mean of EPMC content in bright environmental condition with sucrose concentration 20 g / L medium gave average concentration value of ethyl p-methoxycinnamate 7,49 g / L and with addition of sucrose 30 - 40 g / L an increase of 2.31 to 2,39 times While the environmental treatment without light (dark) with a sucrose concentration of 20 g / L medium of 3.21 g / L, with sucrose concentration added up to 30-40 g / L there was an increase of ethyl p-methoxycinnamate levels ranging from 1.18 to 2.01 times
