- Abstract
Leucocytozoonosis is caused by Leucocytozoon caulleryi and responsible for death in chicken by bleeding. Leucocytozoonosis is an endemic disease in Indonesia and the incidents had been reported in several region in East and Central Java. The financial losses impacted by this disease is growth disorder in chick, decreased egg production, higher mortality rate and also higher production cost. This research aims are to detect TLR-2 and CD4 expression as cellular immune response in rabbits immunized by protein of L.caulleryi schizont. It is needed a preliminary research for molecular vaccine development is considerably effective to prevent the leucocytozoonosis occurance in Indonesia. This research was performed in several stages i.e. isolation of L.caulleryi schizont from chicken liver infected by leucocytozoonosis to be based on clinical signs observed, microscopic examination, and pathological changes of other chicken organs; purification of soluble protein L.caulleryi schizont; immunization of rabbits and each of experimental rabbits injected by 500 æg L.caulleryi schizont protein and added adjuvant complete with ratio 1:1. Every two weeks the injection was performed with the same protein with the dosage 500 æg each and added adjuvant incomplete (booster was performed 5 times in 2 weeks). The examination of cellular immune response of CD4 and TLR-2 expression in rabbits T cells using immunocytochemistry method visualized by fluorescein isothiocyanate. The examination result by immunocytochemistry shows TLR-2 and CD4 expression are yellow to green fluorescence color, mainly in the 5th booster where the activation of CD4 co-receptor and TLR-2 was occured. Conclusion shown that antigenic protein of L.caulleryi schizont has ability to induce cellular immune response through co-receptors CD4 and TLR-2 in rabbits T cells and as preliminary research in sub-unit vaccine development for leucocytozoonosis on chicken.
